Dr. Madoff earned his Ph.D. in a joint M.D.-Ph.D. program at the University of Medicine and Dentistry-Robert Wood Johnson Medical School working in the lab of Dr. John Lenard. He studied the assembly and biochemistry of LaCrosse virus, a Bunyavirus that is a known cause of meningitis and encephalitis.
Dr. Madoff worked as a postdoctoral fellow in the lab of Dr. M. Daniel Lane, then Chairman of the Department of Biochemistry at the Johns Hopkins University School of Medicine. Dr. Madoff studied the basic biochemistry of insulin action.
